Performance Comparison
Comparison of performance with products of other companies in fed-batch culture using each cell line
CHO-DG44
“BASAL3” and “FEED2” were evaluated, compared with eight commercially available culture media in fed-batch culture using a DG-44 cell line.
The model cell was adapted to each basal medium for 3 passages before evaluation.
5% of “FEED2” was added on days 4, 7, 9 and 11 of the cultivation.
The feeding manners for commercial media sets followed each instruction’s recommendation.
As a result, the Ajinomoto products showed the highest cell proliferation and IgG titer compared with the other products.
CHO-S
The model cell was adapted to each basal medium for 3 passages before evaluation in a flask. Using BASAL3 or BASAL10 (CELLiST™) as basal media, 4% of FEED2 (CELLiST™) as feed media was added on days 4, 6, 8, 10 and 12 of the culture.
For the products of other companies, the combination of five types of basal media and the feed media recommended for each were added in accordance with the instructions for each product.
As results of all of the combinations, our CELLiST™ showed higher IgG titer than the products from other companies.
Cost Comparison
Comparison of the total cost of culturing and the cost per antibody concentration (CHO-S)
Using cells of the CHO-S strain, a comparison was performed of the costs when using our CELLiST™ culture media and when using five types of fed-batch cultivation from other companies.
When the total costs (Basal + Feed) and the cost per 1 g of antibodies (Total cost of each culture medium ÷ Titer for each on final day) are examined,
our culture media can suppress the costs more than the culture media from other companies.
To calculate the cost, we referred to the feed manner described in the instructions for each media, and to the price of lab-scale media in the Japanese market.
